Preparation Procedures:
Materials needed but not provided in the kit:
2-mercaptoethanol
ethanol
1. Cut 1-3 mm of mouse tail or toe tips from mice into 1.5 mL microcentrifuge tubes.
2. Lyse tail biopsy for 4 hours to overnight in 500ul Lysing Solution (with proteinase K and 2- mercaptoethanol).
Add fresh to aliquots of solution:
5ul 2-mercaptoethanol (not included) per 1ml lysing solution
20ul Proteinase K (20mg/ml) per 1ml lysing solution
3. Incubate at 55 oC.
4. Vortex 10 seconds. Add 250ul Salt Solution and vortex 5 seconds. Let stand 15 minutes on ice.
5. Spin 15 min at maximum speed at 4 oC
6. Transfer supernatant to clean tube and precipitate DNA with 750ul ethanol. Mix well. Pellet by spinning gently at room temperature at speed “10” for 2-3 minutes.
7. Wash DNA pellet with 750ul 70% ethanol and air dry. Dissolve in 100ul TE or DNase-free water. Resuspend DNA rapidly by incubating 15 minutes at 55 oC
