[讨论]anybody here do perforated patch&am

I'm just wondering if anybody use this technique here? I'm using gramacidin as pore-forming agent. What do you use?

没用过，不过楼主可以发个贴子介绍一下

gramacidin, amphotericin, nystatin 都可以用， 但每个药物的特性不同。

如果要保持chloride离子浓度不变的话， 用gramacidin。 因为gramicidin pores are  only permeable to Na+ and K+ but not to Cl-.

amphotericin, nystatin 允许chloride 离子通过。 一般都可以用。 但nystatin形成孔更快更有效， 所以我们lab大多不用amphotericin(而且很难溶于pipette solution).

现在大体上还是用amphotericin B的多，主要参考文献是James Rae的这篇文章(不过这个人的研究领域实在是无聊的要命)
