ECL, chemiluminescence detection kit
MacGlu gives you a sensitive and convenient method for rapid detection of proteins in Western Blotting.
Description:
MacGlu is a chemiluminescent system for detection of horseradish peroxidase (HRP) labeled antibody on Western blotting. The system combines a reagent that keeps strong and long-lasting chemiluminescent signal.
MacGlu comes in two 100ml color coded bottles. The reagents are stable at 4°C for 1 year*. Due to the sensitivity of the system, and as with all chemiluminescent systems, it is critical to optimize the conditions of your assay, particularly the concentration of primary and secondary antibody to obtain an optimal signal-to-background ratio.
Precedure:
1. After the final wash, place your membrane onto absorbent paper.
2. For 10x10cm membrane, mix 1 mL of A and B in a glass dish.
3. Incubate the membrane (facing down) to ensure even coverage.
4. Touch the membrane against absorbent paper before transfer it into a plastic wrap**.
5. Place the wrapped membrane (face up) in a film cassette
6. Expose to X-ray film for varying amounts of time***.
Note:
*do not expose the solution in the air for a long time, which could affect the sensitivity of the reagents.
**being careful to remove any air bubbles between the membrane and the wrap.
***we suggest you try 15 second, 1 minute, and 5 minutes.
